Here, we suggest to use microscopy imaging to assess the membrane tension. We report direct dimension of membrane layer nanotube diameters with unprecedented resolution making use of stimulated emission exhaustion (STED) microscopy. For this function, we incorporated an optical tweezers setup in a commercial microscope prepared for STED imaging and established micropipette aspiration of huge vesicles.
