Arginine auxotrophy due to the silencing of argininosuccinate synthetase 1 (ASS1) occurs in many carcinomas and in the majority of sarcomas. Arginine deiminase (ADI-PEG2 therapy exploits this metabolic vulnerability by depleting extracellular arginine, causing arginine starvation.
